Plerixafor is an inhibitor of the CXCR4 chemokine receptor and blocks the binding of its homologous ligand, stromal cell-derived factor-1 α (SDF-1α). Sdf-1 α and CXCR4 are thought to play a role in the transport and homing of human hematopoietic stem cells (HSC) to the bone marrow compartment. Once in the bone marrow, stem cell CXCR4 can help anchor these cells to the bone marrow stroma either by sdF-1 α or by inducing other adhesion molecules. Treatment with plerixafor resulted in leukocytosis and an increase in circulating hematopoietic progenitor cells in mice, dogs and humans. CD34+ cells mobilized by Plerixafor were able to be implanted in a canine transplant model for up to one year.
